چکیده
Since the demonstrated clinical success of BRAF/MEK inhibitors in patients with metastatic melanoma harboring BRAF mutations, attention has been focused on liquid biopsy to detect mutations cell-free DNA (cfDNA) using droplet digital PCR (ddPCR) or next-generation sequencing methods. In this study, we investigated gene tumor and cfDNA collected from 43 evaluate their potential as biomarkers. ddPCR CAncer Personalized Profiling by deep Sequencing (CAPP-Seq) techniques were performed plasma obtained melanoma. The sensitivity specificity for mutation detection 54.5 % 100 %, respectively. Additionally, variants, including NRAS, TP53, GNAS, MET, detectable cfDNA, results partially consistent those identified tissues. Among variants examined, copy numbers MET disease status two treated a inhibitor. conclusion, CAPP-Seq presence especially when tissue biopsies are unavailable. may be biomarker

Multiparametric Analysis of Cell-Free DNA in Melanoma Patients
Cell-free DNA in blood (cfDNA) represents a promising biomarker for cancer diagnosis. Total cfDNA concentration showed a scarce discriminatory power between patients and controls. A higher specificity in cancer diagnosis can be achieved by detecting tumor specific alterations in cfDNA, such as DNA integrity, genetic and epigenetic modifications. The aim of the present study was to identify a seq...

Cell-free circulating tumor DNA in the plasma of cancer patients has become a common point of interest as indicator of therapy options and treatment response in clinical cancer research. Especially patient- and tumor-specific single nucleotide variants that accurately distinguish tumor DNA from wild type DNA are promising targets. The reliable detection and quantification of these single-base D...

متن کاملCell-free DNA detected by “liquid biopsy” as a potential prognostic biomarker in early breast cancer
As conventional biomarkers for defining breast cancer (BC) subtypes are not always capable of predicting prognosis, search for new biomarkers which can be easily detected by liquid biopsy is ongoing. It has long been known that cell-free DNA (CF-DNA) could be a promising diagnostic and prognostic marker in different tumor types, although its prognostic value in BC is yet to be confirmed. This r...
